The world number one opened up with a total clearance of 145 and finished his demolition job with a 146 to win in just 64 minutes.
Vafaei, who is playing through the pain of a spinal problem, scored just 31 points in the match.
Trump set up a semi-final with Stuart Bingham, who came from 4-2 down to beat Mark Selby 5-4.
Shaun Murphy knocked out veteran Scot John Higgins 5-3 and will face Neil Robertson, who beat Xiao Guodong by the same scoreline, in the last four.
